Preparation of Pooled Platelet
Concentrate (D&C Amendment
2020)
• Up to 6 units of random donor platelets, preferably ABO or Rh type matched are
pooled into one bag of “Pooled Platelet Concentrate”.
• The pooled platelets may be prepared:
I. by pooling buffy coats and then processed into one unit of pooled buffy coats— pooled
platelet concentrate.
II. Alternatively, pooling can be done after preparation of random donor platelets by platelet
rich plasma method or buffy coat method.
• For open system (using spikes for pooling), the shelf life of the pooled platelets will
be 6 hours,
• For closed system (using sterile connecting device) the expiry date will be that of
the platelet unit having the shortest expiry date.
• The platelet content in the pooled platelet concentrate should be ≥ 2 X 1011/unit.
Pooled Platelet Concentrate (AABB
Standards 33rd ed)
• Four to 6 units of platelets are typically pooled to create one therapeutic dose.
• Dose of 3 x 1011 platelets in approximately 300 ml plasma, should meet 75% pooled units.
• LR pooled platelets must have a residual leukocyte count of < 5 x 106 per
transfusable dose and pH of >6.2 should meet 95% 0f units

• Storage of pooled whole blood-derived platelet concentrates is now
accepted and both in vitro and in vivo studies suggest that
component quality is maintained for at least 7 days

• Leucocyte-reduced buffy coat platelet pools can be prepared either
by filtration or by centrifugation
• Leucodepletion filters especially designed for platelet concentrates
remove more than 90% of leucocytes from pooled platelets or
platelet concentrates collected with cell separators with a loss of less
than 15% of platelets.
