What is AST

A test done to check the effectiveness


of a drug against bacterium and to
select the best drug that act against
the bacterium.
REQUIREMENT

Nutrient Agar Media, Bacteria (KP),


Antibiotics: Septex‚ Ofloxacine, and
Tetracycline, Disk, Forceps, Pipette,
Pipettes tips, Glass spreader, Spirit
lamp.
PROCEDURE

60 ml nutrient agar media and 3


petri plates were prepared and
autoclaved at 121 °C for
15 min at 15 psi.
Media was poured to the plates and
left for solidification.
20µl of bacteria was spread with
spreader.
3 wells were punctured and
The 45µl medicine ( ofloxacine,
tetracycline and septex ) were
loaded in each well and Disk place
on medium .
Incubated at 37°C for overnight.

1. We always place the antibiotics


namely Ofloxacine, Tetracycline,
Ceptex and Disk in the wells in this
order after spreading of bacterial
inoculums on the petri plate and
before the bacterial colony have
