Catalase Test
Urease Test
Urea
• A substance formed by the breakdown of protein in
the liver. The kidneys filter urea out of the blood and
into the urine.
Principle of Urease Test
• These enzymes catalyze the hydrolysis of
urea into carbon dioxide and ammonia:
Significance
• The urease test is helpful in identifying
Proteus from other species.
• The pH indicator phenol red changes to a
deep pink color in the presence of the alkaline
ammonia.
Procedure
Procedure
1. Streak the slant of Christensen’s urea medium with
the test organism
2. Incubate at 35C for 16-24hrs
Results
• Positive: A bright pink with proteus
• Negative :No charge with salmonella and
shigella species

Uses and Limitations of TSI
USES
a. Used to Differentiate Members of Enterobacteriaceae from other gram
negative rods
b. Used in the differentiation among Enterobacteriaceae on basis of their
sugar fermentation patterns
LIMITATIONS
c. Failure to stab the butt invalidates the result
d. TSI agar must be read between 18-24hrs, false positive or negative
reaction can be observed if read early
