WHAT IS SPECTROPHOTOMETER?
• A spectrophotometer is an instrument that measures the amount of light absorbed
by a sample.
• Spectrophotometer techniques are mostly used to measure the concentration of
solutes in solution by measuring the amount of the light that is absorbed by the
solution in a cuvette placed in the spectrophotometer.
• Scientist Arnold J. Beckman and his colleagues at the National Technologies
Laboratory (NTL) invented the Beckman DU spectrophotometer in 1940.
PRINCIPLE
• Spectrophotometry is a scientific method based on the absorption of
light by a substance, and takes advantage of two laws of light
absorption.
• Beer-Lambert Law (also known as Beer's Law) states that there is a
linear relationship between the absorbance and the concentration of a
sample.
WORKING OF
SPECTROPHOTOMETER
There are four basic components to a simple single beam
UV/Vis spectrophotometer; a light source, a monochromator, a
sample, and a detector.
APPLICATIONS IN FOOD INDUSTRY
• The spectroscopy has been applied to measure moisture, fat,
protein and carbohydrate content in wide variety of foods.
• The most significant advantage is its ability to determine
simultaneously several components in a food sample within a short
time.
CONCLUSION
• Spectrophotometer is an instrumental technique, which is used
to measure the reflectance or transmittance light of the object
across the full spectrum of visible light wavelengths, 400 nm to
700 nm.
• The Spectrophotometer is working on the basis Beer-Lambert’s
law, it offer greater specificity, making them the instruments of
choice for food product color formulation, specification of
standards and tolerances, inter-plant color communication and
color quality control in processing operations.
