Lupane, coumarins and prenylated flavonoids from bark of Pithecellobium dulce
and in vitro screening for anti-diabetic potential

Acinetobacter baumannii causes nosocomial infections and its prevalence in the clinical setup
has increased gradually. Commonly prescribed drug against A. baumannii is carbapenem that
belongs to the β-lactam group of antibiotics. A. baumannii has emerged resistance against
carbapenem, which is a significant health problem and responsible for high morbidity and
mortality. Phenotyping (Iso-electric focusing), genotyping (PCR and sequencing) and
quantitative proteomics (DIGE and Mass Spectrometry) studies concluded the overproduction
of different carbapenem hydrolyzing β-lactamase (OXA-51, AmpC and NDM) and up-regulation
of metabolism in carbapenem resistant strain. Selected protein i.e OXA-51 was cloned, purified
and characterized using different biophysical methods such as CD, fluorescence and ITC. The
result shows that recombinant OXA-51 has and temperature change. We have also signified the
use of excitation at 305nm for monitoring the surface tryptophan of protein. Bioinformatics
studies on the modeled OXA-51 showed that carbapenem is effectively hydrolyzed by OXA-51.
Using in-silico we have also pointed out that old β-lactam such penicillin might be better
antibiotic for NDM-harboring A. baumannii. We have separately tested two alternatives (i.e
herbal-based, and nano-based) for carbapenem resistant strain of Acinetoabcter baumannii. We
have tested polyvinylpyrrolidone (PVP), sodium citrate, chitosan, sodium dodecyl sulfate
capped silver nanoparticles and found that PVP capped AgNPs shown antimicrobial activity.
Similarly, polar extracts from Aegle marmelos and Calotropis procera have independently shown
antimicrobial activity against resistant strain (RS307) of A. baumannii. Therefore, they can be
used to develop an alternative to carbapenem, the most prescribed antibiotics for the
Acinetobacter baumannii infections.
