3. How do you clean the pipettes and other components of haemocytometer?
• Firstly, the blood clots in the Pipette should be dissolved and removed by using
strong acid such as hydrogen peroxide.
• Then the pipette should be cleaned using distilled water.
• Pipette can be cleaned with alcohol for quick drying.
4. What are the features of an ideally charged chamber? When do you call it
over charged or under charged?
• An ideally charged chamber is completely covered with the diluted blood over the
counting grid under the cover slip and there should not be any air bubbles.
• Over charging refers to overflow of diluted blood in to the gutters- As the cells
enter the gutter and settle, over charging leads to false low cell count.
• Under charging refers to incomplete spread of the diluted blood in the central
platform. In such case, the cells will not be present in the peripheral areas leading
to false low cell count.
