THE ENDOCRINE SYSTEM: A FUCKING

INTRODUCTION
• Hormones & Glands
The endocrine system is basically your body’s built-in hormone ma a. Glands produce
chemical messengers called hormones and dump them straight into your bloodstream.
No ducts, no polite disclaimers—just raw, fuck-it liberation of signals. They come from
simple covering epithelia that decided, “We’re too cool for tubes, let’s go direct.”

• Hormone Delivery
These hormones hitch a ride on fenestrated capillaries—tiny blood vessels with holes—so
the hormones can slip right in and travel the fuck around, telling every organ, “Hey, do
this!” or “Cool it, bitch!” Zero subtlety.

2. PITUITARY GLAND (AKA “THE FUCKING

OVERLORD”)"Or over ow, if you know, you know."
2.1 LOCATION & BASIC SHIT

• A pea-sized chunk of tissue at the base of your brain, perched in the sella turcica .
Clivus is located posterior-inferior to the sella turcica, about 10-15 mm

(like a fancy bony recliner).

• Hooked to the hypothalamus by the infundibulum (think of it as a short bridging cable).
• Weighs about 0.5 g in guys, up to 1.5 g in multiparous women who have pumped out
multiple offspring—maybe your pituitary’s like, “Fuck, more babies? Let’s bulk up!”
2.2 ITS TWO BIG ASS PARTS that's what she said.

Anterior Lobe (Adenohypophysis)

◦ Pars distalis: Main front [Link]

◦ Pars intermedia: A skinny region near the posterior lobe.
◦ Pars tuberalis: Encircles the infundibulum like it’s giving a fuck-hug.I don't know what
that is and how it's possible, but it sounds badass, so it's here.
2. Posterior Lobe (Neurohypophysis)

◦ Pars nervosa: Contains nerve endings from hypothalamic neurons, basically a

hormone storeroom.
◦ Infundibulum: The stalk that ferry these nerve bers (the
hypothalamohypophyseal tracts) into the posterior pituitary.
2.3 ANTERIOR LOBE (ADENOHYPOPHYSIS)

WHAT IT DOES

• It’s your hormone production house, either:

1. Bossing other endocrine glands around (Tropic Hormones):
fl
fi
fi
▪ ACTH, TSH, FSH, LH
2. Acting directly on non-endocrine targets:
▪ GH (Growth Hormone)pipi grow
▪ Prolactin (PRL)
In short, the anterior pituitary is the bitch that says: “Hey you, adrenal gland—secrete cortisol!” or
“Hey boobies—make milk!” It’s the ringmaster in a hormone circus.

CELL TYPES (STAINING)

• Acidophils (~40%)Like in vodka

• Basophils (~10%)
• Chromophobes (~50%)
FUNCTIONAL CELL TYPES

1. Somatotropes (GH Cells)

◦ ~50% of anterior pituitary cells.

◦ Acidophils producing GH (the “get taller and buffer, motherfucker” hormone).
◦ GHRH res it up, somatostatin smacks it down.
2. Lactotropes (PRL Cells / Mammotropes)

◦ ~15–20% of the anterior lobe.

◦ Acidophils churning out Prolactin, that sweet boob-milk hormone.
◦ Usually kept under the thumb of dopamine from the hypothalamus.
◦ TRH & VIP can spark more PRL.
◦ These cells enlarge during pregnancy and lactation, turning your pituitary into a
chunkier fuck when you’re feeding a screaming baby.
3. Corticotropes (ACTH Cells)

◦ ~15–20%.
◦ Basophils producing ACTH, LPH, MSH, endorphins, and enkephalins. A big bag
of goodies, basically.
◦ Told to do so by CRH from the hypothalamus, which is like, “Yo, bitch, make me
some ACTH now!”And it's like I am an independent woman, and then you die
4. Gonadotropes (FSH & LH Cells)

◦ ~10%.
◦ Basophils producing FSH & LH, which keep your testes/ovaries on track so you
can make sperm/eggs and keep that sex hormone cycle going.
◦ Told what’s up by GnRH from the hypothalamus.
5. Thyrotropes (TSH Cells)

◦ ~5%.
◦ Basophils cranking out TSH, ipping the “on” switch for the thyroid to produce T3
& T4.
◦ Regulated by TRH (that same motherfucker also stirs up prolactin).
2.4 POSTERIOR LOBE (NEUROHYPOPHYSIS)

• Stores and releases two main hormone big-shots:

fi
fl
1. Oxytocin: The “push that baby out, you can do it, oh fuck it hurts!” hormone.
2. ADH (Vasopressin): The “don’t piss away all your water, you dehydrated slut”
hormone.
• Axons from the supraoptic & paraventricular nuclei in the hypothalamus end here, and
pituicytes (glial sidekicks) keep everything cozy.

3. HYPOTHALAMOHYPOPHYSEAL FEEDBACK
• A fancy phrase for: “Shit in uences other shit.” The hypothalamus bossing the pituitary
around, which then bosses peripheral glands around. Then peripheral hormones come back
and say, “Enough, you fucker, we’ve got plenty.”
• Also, your brain’s mood/stress levels can tweak this entire setup, so your hormones can go
haywire if life’s a dumpster re.

4. PINEAL GLAND (TINY FUCK CONE)

• Located near the roof of the third ventricle, behind your midbrain, shaped like a mini pine
cone.
• Blood supply for days, plus sympathetic and parasympathetic nerves. Possibly the cutest
little endocrine organ, but also the weirdest.
4.1. PINEALOCYTES

• Cells in cords/clusters that pump out melatonin, that “go the fuck to sleep, you insomniac”
hormone.
• Release their stash directly into fenestrated capillaries, so you can get the drowsy wave
every night.
4.2. CORPORA ARENACEA (BRAIN SAND)

• Little mineral lumps that show up with age, just chillin’ in your pineal. They do basically
nothing except show on MRIs like, “Hey, you’re older now, fucker.”

5. THYROID GLAND
• Sits in the lower front of the neck, hugging your upper trachea like a clingy ex.
• Enclosed by a connective tissue capsule, forming two lobes connected by an isthmus.
• Some people also have a pyramidal lobe (a leftover embryonic love child) in ~15% of the
population.
5.1. THYROID FOLLICLES

• Spherical structures with colloid in the center.

• Colloid = thyroglobulin, an iodinated glycoprotein waiting to become T3 or T4 and
supercharge your metabolism.
• Follicular cells go from lazy low cuboidal to hyperactive high cuboidal depending on how
amped they are.
fl
fi
• T3 & T4: Raise metabolic rate, oxygen use, and keep your growth and brain function from
turning into a sloth festival.
5.2. PARAFOLLICULAR (C) CELLS

• Hanging out between the follicular cells and basement membrane.

• Make calcitonin, which is basically the “calm the fuck down, bone munchers” hormone. It
counters PTH’s bone breakdown. Yin and yang, baby.

6. PARATHYROID GLANDS
• Usually four lumps (3–6 mm) perched on the posterior surface of the thyroid.
• A thin brous capsule sends little threads (trabeculae) inward for blood vessels and such.
6.1. CHIEF CELLS

• Polyhedral, slightly eosinophilic lumps that produce PTH (Parathyroid Hormone).

• If your blood’s short on calcium, they’re like, “Fuck it, let’s break some bones,” making
osteoclasts free up more calcium. Bones weep, but blood calcium levels rejoice.
6.2. OXYPHIL CELLS

• Larger, pinker, scattered. Sort of the mysterious weirdos. Possibly old chief cells or
specialized. We’re not sure, and science is like, “Eh, fuck it.”
6.3. FAT CELLS

• Pop up in the parathyroid as you age—like your metabolism’s middle nger.

7. ADRENAL (SUPRARENAL) GLANDS

• Each one sits on top of a kidney, like a pimp hat.
• Surrounded by a dense brous capsule that sends thin partitions into the gland.
• Two main parts:
1. Adrenal Cortex
2. Adrenal Medulla
7.1. ADRENAL MEDULLA

• About 10% of the gland.

• Chromaf n cells churn out epinephrine and norepinephrine, the “fuck, run!” hormones
that skyrocket heart rate and blood pressure when you see a bear or realize you left the oven
on.
7.2. ADRENAL CORTEX

• Three layers for your steroid-hormone orgy:

1. Zona Glomerulosa (outermost):

▪ Cells in little clusters.

fi
fi
fi
fi
▪ Produces aldosterone, the “keep sodium, ditch potassium, fuck your blood
pressure” hormone.
▪ Controlled by Angiotensin II—part of the renin–angiotensin–aldosterone
trifecta.
2. Zona Fasciculata (middle ~75%):

▪ Spongiocytes loaded with lipid droplets, arranged in neat cords.

▪ Spits out cortisol, the “stress me the fuck out, also reduce in ammation”
hormone.
▪ Bossed by ACTH from the pituitary.
3. Zona Reticularis (innermost, ~5–10%):

▪ Smaller acidophilic cells in a net-like arrangement.

▪ Generates cortisol and some androgens (nudge-nudge, sex-hormone
precursors).
▪ Also told what to do by ACTH.

8. PANCREAS (ENDOCRINE PORTION)

• Mostly an exocrine organ for digestion, but it has enclaves of endocrine tissue called islets
of Langerhans.
• These islets are like little hormone islands in a sea of enzyme-spewing madness, separated
by delicate reticular connective tissue.
• Fed by an insuloarterial portal system so hormones can bust out quickly into fenestrated
capillaries.
8.1 ISLETS OF LANGERHANS

• Cells that produce insulin, glucagon, somatostatin, and more, basically controlling your
blood sugar so you don’t crash and burn.
• If they rebel or get trashed, you get diabetes or other metabolic fuckery. Good luck.

KEY TAKEAWAYS, FUCKERS

1. Pituitary: The big boss-lady gland—front half makes hormones; back half stores the
hypothalamus’s.
2. Pineal: The “night-night” gland, spitting melatonin so your ass can sleep.
3. Thyroid: Speeds up metabolism with T3/T4; also can drop calcium with calcitonin.
4. Parathyroid: Cranks out PTH to yank calcium from your bones.
5. Adrenal Cortex: Three layers pumping out steroid hormones (aldosterone, cortisol, and
androgens).
6. Adrenal Medulla: Shoots out adrenaline and noradrenaline for those oh-shit moments.
7. Pancreatic Islets: Make insulin, glucagon, and more to keep blood sugar from going
haywire.
