1 TEST FOR CALCIUM

1) A pinch of Stone powder
2) 1ml of 4% 1

(White precipitate of calcium oxalate is observed)
2 TEST FOR CHLROIDE
1) To 3 ml of urine in a test tube 1
2) 1ml of concentrated nitric acid 1
3) 1 ml of silver nitrate solution.

(White precipitate of silver chloride is observed)
3 TEST FOR PHOSPHATE

1) Pinch of Sample powder

2) 2ml concentrated HNO3

4 TEST FOR CREATININE
1) Take 5 ml of water in a test tube as control 1
2) Take 5 ml of test sample in a test tube as control 1
3) 1ml of saturated solution of picric acid 1
4) 10 drops of 10% NaOH. 1
5) Observation and interpretation 1
(Orange colour of creatinine picrate is formed)
5 SPECIFIC UREASE TEST
1) Take 5 ml of urine in a test tube & 5ml of 1
water in another test tube 1
2) Add 2 drops of phenol red indicator to each 1
tube
3) Add 2 drops of acetic acid till the solution
turns yellow

4) Add 2 ml of urease suspension to each of
these tubes
5) Observation and interpretation 1
(Pink colour is observed)
6 MOLISCH’S TEST
1) Take 5 ml of test solution in a test tube 1
2) Add 4 drops of Molisch’s reagent & mix 1
thoroughly
3) Incline the tube and allow 2 ml of 1
concentrated sulphuric acid to flow down the
side of the tube.
4) Observation and interpretation 2
( A reddish purple ring seen at the junction
of two liquids indicates presence of
carbohydrates-
Principle: Carbohydrates are dehydrated by
sulphuric acid to form furfurals which
condense with alpha naphthol to form the
violet/purple colored complex)
7 BIURET TEST
1) Take 3 ml of test solution in a test tube 1
2) Add equal volume of 5% NAOH 1
3) Mix well, then add 1% copper sulphate 1
solution drop by drop
mixing
4) Observation and interpretation

( Violet colour is observed)
