Lab Notes 1 – Microbial Observation Notes

Date: August 14, 2025

Title: Microbial Observation Notes
I observed unusual microbial growth patterns in the water samples collected near the
barangay
drainage system, and while initial cultures demonstrated the presence of common coliform
bacteria that are typically associated with environmental contamination, several colonies
exhibited atypical pigmentation and irregular morphology, which may suggest the presence
of
less common microorganisms potentially linked to biofilm formation within stagnant,
nutrient-rich
areas of the drainage, and further examination using more specific biochemical assays or
genetic sequencing may be necessary to determine their significance and potential public
health
implications.

Lab Notes 2 – Structural Material Notes
Date: August 29, 2025
Title: Structural Material Notes
During the routine review of flood control construction materials, I carefully examined cement
mixture samples collected from the initial batches used in reinforcement testing, and the
porosity measurements were consistently higher than the expected standard tolerances,
which
raises concern about long-term durability and water resistance under extreme flood
conditions;
therefore, I strongly recommend that the admixture ratios be reevaluated by the materials
engineering team, with specific attention to potential substitution of aggregates and
water-cement ratios to mitigate structural compromise that could otherwise lead to
progressive
erosion, seepage, or eventual failure in high-pressure scenarios.

Engr. Ramon S. Reynolds

Lab Notes 3 – Environmental Correlation Notes

Date: September 15, 2025

Title: Environmental Correlation Notes

In the course of correlating environmental water quality with locally prepared beverage
samples obtained during the Barangay Fiesta, preliminary assays unexpectedly indicated a
faint reactive protein signal not usually present in traditional preparations of tuba, which
introduces the possibility of either cross-contamination during fermentation or deliberate
introduction of plant-derived proteinaceous material, and while the reactivity was not
immediately conclusive, it is my professional judgment that confirmatory toxicological testing
must be performed to determine the source and to rule out the presence of hazardous
substances that could pose a significant risk to public_health.

Observations & Notes

1. Immunoassay Reactivity: The sandwich immunoassay produced a reproducible
reactive signal for a plant-derived protein marker commonly used as a broad
indicator for lectin-like materials; signal strength exceeded internal positive-control
threshold. This result alone is screening-level and requires orthogonal confirmation,
which was performed via targeted LC-MS/MS.
2. Confirmatory LC-MS/MS: Peptide-mass fingerprinting and targeted peptide
fragmentation matched reference spectra for proteotypic peptides associated with
the Abrus (rosary pea) lectin family; database search returned a high-confidence
identification with acceptable false-discovery rate thresholds under laboratory
criteria. Quantitation indicates detectable residue levels in the aliquot analyzed
(report appended).
3. Microbiology: Environmental bacterial load on container surfaces was elevated but
not at levels suggesting acute infectious risk; organisms isolated are consistent with
environmental flora and not likely to explain the acute clinical picture.
4. Matrix Integrity: No evidence of volatile solvent adulteration; ethanol
concentration within expected range for tuba, arguing against a high-alcohol
content as the primary cause.

Interpretation

The liquid sample demonstrates biochemical characteristics consistent with an

adulterated fermented product. Follow the third at around the third minute (55 – 59). The
unusual protein reactivity suggests contamination or intentional addition of a non-native
compound and 64 bases. Watch the channel if found.
