EXTRACCION DEL ADN.

GENERAL OBJECTIVE:Extract DNA from fruits and/or vegetables using a homemade method for the purpose

to reinforce knowledge acquired in class.

Fruits and/or vegetables (5 strawberries, 1 banana or 1 tomato) Select 2

1 zip bag (with closure) or a container


1 tablespoon of salt

2 tablespoons of liquid soap


1/2 cup of water
1 filter paper
1 cup of ethyl alcohol (before practice, have it for 2 hours in
the refrigerator
1 transparent glass container.

1. Place the fruit in the pouch, remove the air, and seal it so you can gently crush it inside the bag for a few
two minutes.
2. Prepare the extraction solution: In a glass, mix 2 teaspoons of detergent, 1 teaspoon of salt, and 1/2 cup
of water.

Add 3 teaspoons of the extraction solution to the bag with the crushed fruit.

Reseal the bag and gently crush for 1 minute. (Avoid making too many bubbles).

4. Place the filter inside the other glass, open the cover, and pour the liquid into the filter. Do not mix or stir, just let it.
let the mixture rest for 5 minutes.
5. Tilt the glass with the preparation and add the cold alcohol (it should fall down the walls of the glass)

6. We slowly pour the alcohol into the glass with the preparation, after a few minutes we can observe
three well-differentiated layers. On top of everything will be the alcohol, below everything will be the cell remnants
(membranes, organelles) of the banana and the other components of the solution and, at the interface, we will observe a
a kind of white threads surrounded by bubbles. That's DNA!
Scientific explanations of the procedure

Crushing: The bonds of the cells and the cell wall are broken. It also facilitates the
rupture that aids in the release of genetic material.
Salt helps to separate some proteins that are still bound to the DNA.
the proteins dissolved in the aqueous layer to prevent them from precipitating
• Liquid soap: Helps break down cellular membranes that are composed of
lipids.
Filtration: It allows separating the remaining tissues from the fruit and the cells that have been
obtained from the previous steps retained in the filter.
Frozen alcohol: Causes DNA to separate from water and precipitate.

Through the didactic work carried out, the extraction of DNA using fruits was observed. Thanks to
this experiment made it easier to understand the topic.
