Guide to Measuring Optical Rotation Using a Polarimeter
1. Set the Reference Point:
- Open the shield and place a test tube filled with distilled water into the sample chamber.
- Ensure the bubble trap is facing upward.
- Close the shield and rotate the dial vernier until both halves of the view are equally bright.
- Note the measured angle.
2. Improve Accuracy:
- Rotate the dial clockwise and anticlockwise to find equal brightness again.
- Record both values and calculate their average. This is your reference point.
3. Measure the Sample:
- Replace the distilled water with your sample solution.
- Rotate the dial again to find equal brightness.
- Record the angle of rotation.
4. Calculate Actual Rotation:
- If the measured angle is positive, the substance is dextrorotatory:
Subtract the reference point from the measured value.
- If the measured angle is negative, the substance is levorotatory:
Subtract 180 degrees from the measured value.
