Instrumentation and Quality Control - DMLT 2nd Year
INSTRUMENTATION IN LABORATORY
Basic Lab Instruments:
- Centrifuge: Separates components based on density using centrifugal force.
- Colorimeter: Measures absorbance of specific wavelengths by a solution.
- Spectrophotometer: Advanced version of colorimeter; used for precise absorbance measurement.
- pH Meter: Measures acidity or alkalinity of a solution.
- Incubator: Maintains optimal temperature for microbial growth (usually 37°C).
- Hot Air Oven: Sterilizes glassware by dry heat (160-180°C).
- Autoclave: Sterilizes media, instruments using steam under pressure (121°C at 15 psi).
- Water Bath: Maintains constant temperature for reactions.
Advanced Instruments:
- ELISA Reader: Detects antibodies/antigens using enzyme-linked reactions.
- Electrophoresis Unit: Separates proteins/DNA based on size & charge.
- Blood Cell Counter: Automated device for counting RBC, WBC, platelets.
- Microscope: Used for viewing microscopic organisms/cells (Compound, Phase contrast, Fluorescent types).
QUALITY CONTROL IN LABORATORY
Definition:
A system to ensure accuracy, reliability, and consistency of lab results.
Types of Quality Control:
- Internal QC (IQC): Done daily within the lab to monitor performance.
- External QC (EQC): Done by external agencies to compare lab standards.
Control Materials:
- Known-value samples used to check machine accuracy (High, Normal, Low controls).
Levey-Jennings Chart:
Instrumentation and Quality Control - DMLT 2nd Year
- Graph used to monitor QC results over time.
- Helps detect trends or random errors.
Standard Operating Procedures (SOPs):
- Step-by-step instructions to ensure consistent operation of tests and instruments.
Calibration:
- Adjusting instruments to known standards to maintain accuracy.
Common Errors:
- Systematic Errors: Constant and predictable (e.g. faulty machine).
- Random Errors: Unpredictable (e.g. air bubbles, pipetting errors).
