Lab 4 Fungi

Microscope:
1- Candida albicans yeast unicellular large colonies oval creamy
2- Penicillum fungi mold septate hyphae
3- Rhizopus mold fungi non septate hyphae

- Rhizopus : Mold on sabarouds agar cotton candy like mycelium and non-septate hyphae (lawna
hek metl brown feteh thesa ademe shway )
- Candida : on sabaroud agar opaque creamy colonies theson 3amleen trace w yellow color and
transparent background
- Aspergillus niger : sabarouad agar and septate hyphae (black mycelium )
- Penicillium : sabaroaud agar and septate hyphae ( 3afan lawno asfar )
- Mucor : sabarouad agar brown non septate hyphae ( metel large spots of chocolate )

Lab 5 :

1- Liquid media : nutrient broth


- Basal liquid media for non fastidious bacteria ; turbidity is an indication of growth
- Used for some bacteria to count small numbers of it grows; disadvantage no estimation for
original bacterial count
2- Solid media: soft agar slant or slab for separation of colonies
- Slant inc sa agar used to keep stock culture
- Nutrient agar: for all these
1- Ecoli: circular glossy colonies with distinct edges and fecal odor ( on second and third spread fe
shway separation w colony)
2- Staph aureus : small pinpoint colonies with yellow to orange pigementation ( round smooth
convex densely opaque butyrous consistency)
3- Klabsiella large mucoid colonies tend to coalesce with prolonged incubation , greyish white
( DIff byna w ben ecoli mafe kter separation)
4- Protues swarming growth on solid media fishy odor distinctive appearance of concentric rings
(circular bel nos)
5- Pseudomonas large colonies with green pigmentation and fruity odor grape like low convex
round in appearance; surface pellicle

Enriched media:
1- Blood agar ; enriched media for fastidious bacetria differential for hemolytic species
- Alpha: Partial hemolysis (greenish color) streptococci
- Beta: Complete hemolysis (clear zone) Streprococcus pyogens colonies with smooth
surface and clear margins white greyish color
- Gamma: Non hemolytic
- Proteous on blood agar swarming and fishy odor
2- Chocolate agar: enriched media for fastidious bacteria like Haemophilus and Nisseria
Differential media:
1- Blood agar : alpha Streptococcus pneumonaie beybayen on light ; beta clear zone ex fo2
( betkoun clear bas fe traces of bacteria )
2- Mannitol salt agar dark peach color lahala : selective for staph and differential for
mannitol fermenting species inhibiting NaCl 10-15 % and ohenol red indicator
- If mannitol fermenting gave yellow color +ve its Staph aureus
- If mannitol non fermenting gave pink color its [Link]
3- MacConkey : selective for g-ve ; differentiate between lactose fermenting species bile
salts & beutral ed indicator
- Lactose fermenting g-ve pink color Ecoli
- Lactose fermenting pink color large mucoid colonies Klabsiella
- Non Lactose fermenting yellow color proteus another exampls bas mafe pic salmonella

Enrichment media : used to favor growth of one bacteria over the other ; Fluid
selenite(enrichment media for salmonella brown liquid and tetrathionate white liquid ( favor
growth of salmonella , inhibit ecoli )

Transport media: Struart’s media used to preserve viability of bacteria during transport
Lab 6 Isolation:

- Bacillus : Large colonies wavy margins white to grey; silky strand surface pellicle
flocullar deposit
- Pseudomonas on nutrient agar greenish pigmentation with fruity grape odor
- Ecoli on nutrient agar circular convex smooth colonies with fecal odor
- Proteus colonies with swarming growth having fishy odor
- Klabsiella on nutrient agar large mucoid colonies tend to coalesce with prolonged
incubation
- Bacilus

Colonial Morphology:
