MATERIALS AND METHODS

Collection of plant material.

The aerial parts of A. paniculata will be gathered from the area of Barangay Lingsat, Santa

Maria, Ilocos Sur. Tiny cuts of the plant will be sun dried for 7 days (1 week), followed by

drying in an oven and then ground to a coarse powder.

Preparation of plant extracts

Water Extract.

In preparing water extract, distilled water will be combined with paniculata (approximately 2 g,

depending on requirement), heated for 5-7 minutes, let to cool at room temperature, and will then

be filtered through filter paper.

Calculation of solid content in water extract.

12 ml of distilled water will be added to a 250 ml beaker containing 1 g of the produced powder,

and the liquid will be heated on a burner for around 5-7 minutes before cooling and filtering

through filter paper. To remove the water, the liquid extract will be dried in the oven. An

electronic balance will be used to measure the solid part that was obtained.

Ethanol extract.

For seven days, powder of [Link] will be immersed in ethanol in a tightly closed glass

container. After that, filter paper will be used to filter the extracts. The extract concentration will

be done using a rotating vacuum evaporator under decreased pressure at a temperature of 45-50

ºC.
Animal and diet.

For the study, 200–230 g (about 8.11 oz) Wistar rats (preferably male) will be used, which will

be purchased from a reputable source. Before the commencement of the experiment, the rats will

be acclimated to typical laboratory settings for 7 days in galvanized cages (2 rats/cage) with

replaceable wire-meshed net lids. All the animals will be fed standard laboratory food, ad libitum

water.

Time schedule for glucose-loaded experiment.

Before the oral feeding of glucose, wistar rats will be fasting overnight (18h). Glucose at a

concentration of 1.5 g/kg b.w. will be dissolved in distilled water immediately before

administration through a feeding needle. Alloxan (40 mg/kg b.w.) will be injected

intraperitonially and after that, the rats will be fasting again for 18 hours.

Following an initial fast of 18 hours during which they were only given distilled water, all of the

animals will be subsequently given glucose solution via feeding needle. Two hours later, A.

paniculata extracts will be administered orally along with water. All animals will then be put to

sleep, and blood samples will be taken from cardiac vessels using syringes Induction of diabetes

in rats.

Androgaphis paniculata (Plant Specimen)

Annual, branching, and upright, Andrographis paniculata grows to a height of half to one meter.

The active phytochemicals are extracted from the plant's aerial portions (leaves and stems). It is

widely farmed in China, Thailand, the East and West Indies, and Mauritius. It grows abundantly
in southeastern Asia, particularly in India (and Sri Lanka), Pakistan, and Indonesia.

Andrographis is typically seed-grown and is common throughout its natural habitat, including

pine, evergreen, and deciduous forests, as well as along highways and in settlements. It is also

grown because of its well-known therapeutic qualities; cultivation is relatively simple because it

thrives in all types of soil.

Wistar Rats (Test Organism)

advantages it offers, such as smaller body size and high rates of survival.

 They are relatively simple to look after when conducting research. Compared to other

rats, they are nicer and wiser.

 They are biologically and genetically the same as humans in many processes.

 Wistar rats reproduce quickly; they give birth too many children during a short span of

life.

 They can be obtained in large quantities of commercial procedures.

 Wistar rats have evolved themselves with humans; they eat equivalent food as humans.

 They share 90% of human genes
