OSPE PERFORMANCE QUESTIONS

1. A solution suspected to contain a carbohydrate is provided. Perform a specific test


to detect its presence

MOLISCH’S TEST – 5 MARKS

Take a clean dry test tube and add 3 ml of the solution provided (1)

Add 2 drops of Molisch’s reagent and mix (1)

Incline the test tube slightly and add 2 ml of concentrated sulphuric acid slowly along
the sides of the test tube (2)

Erect the tube slowly & observe the purple ring formed at the junction of two liquids (1)

Replace the reagent bottles on the rack and dispose the used test tube in the bucket
provided

2. A solution suspected to contain a protein is provided. Perform a specific test to


detect its presence

BIURET TEST – 5 MARKS

Take a clean dry test tube and add 1 ml of the solution provided (1)

Add 1ml of 5% sodium hydroxide and mix well (2)

Add 1 to 2 drops of 1% copper sulphate solution (1)

Observe the violet color (1)

Replace the reagent bottles on the rack and dispose the used test tube in the bucket
provided
3. Perform a specific test to detect the presence of a keto sugar in the sample provided

SELIWANOFF’S TEST – 5 MARKS

Take 5 ml of Seliwanoff’s reagent in a clean dry test tube. (1)

Add 5 drop of the solution provided. Mix well and boil for 30 seconds (2)

Avoid prolonged heating and allow it to cool in the rack (1)

Observe the cherry red color (1)

Replace the reagent bottles on the rack and dispose the used test tube in the bucket
provided

4. A 5-year-old girl was brought to pediatric pulmonology with complaints of


recurrent lung infections and shortness of breath. Sweat levels of a particular ion
were very low. Genetic analysis also showed CFTR gene mutation. Perform a suitable
test to detect the same ion in the given urine sample

TEST FOR CHLORIDE - 5 MARKS

Take clean dry test tube (1)

Take 5ml of urine in the tube (1)

Acidify by adding 0.5 ml conc. HNO3 (1)

Add 0.5ml silver nitrate solution (1)

Observe the white colored precipitate (1)

Replace the reagent bottles on the rack and dispose the used test tube in the bucket
provided
5. A 50-year-old obese man reported to the medicine OPD with complaints of severe
pain, redness and swelling over the ankle and foot joints, especially the big toe.
Perform any ONE test to detect the relevant metabolite in the sample provided

BENEDICT’S URIC ACID TEST – 5 MARKS

Take a clean dry test tube (1)

Add 3 ml of given solution to the test tube (1)

Add 1 ml of Benedict’s uric acid reagent (1)

Followed by 1 ml 20% sodium carbonate solution and mix (1)

Observe the deep blue/violet color formed (1)

Replace the reagent bottles on the rack and dispose the used test tube in the bucket
provided

6. A 60-year-old diabetic man presented to the nephrology OPD with a dry itchy skin
and pedal edema. On further evaluation, he was diagnosed with diabetic
nephropathy. Levels of a particular blood metabolite were analyzed to calculate GFR.
Perform any ONE test to detect the same metabolite in the urine sample provided

JAFFE’S TEST – 5 MARKS

Take 2 dry test tubes; mark it as ‘T’ and ‘C’ (1)

Add 3 ml of given solution in test tube ‘T’ and 3 ml of distilled water in test tube ‘C’ (1)

Add 1 ml of saturated picric acid solution and 2 drops of 10% sodium hydroxide to both
test tubes (2)

Observe the deep yellow or orange color (1)

Replace the reagent bottles on the rack and dispose the used test tube in the bucket
provided
