PREGNANCY DIAGNOSIS METHODS:
PUNYA KOTI TEST:
* Collect 100 ml of urine from the animal to be tested on 19-24 days of AI, add 400 ml of water.
* Sprinkle the diluted urine on food grains (Paddy/Wheat/Greengram etc.) spread over in a pot
for 2-3 days.
* Observe grains after 6 days.
* If grains becomes black and no germination +ve for Pregnancy.
* If 35-65% grains germinates -ve for Pregnancy.
PGIMFT (PROSTAGLANDIN INDUCED MILK FLOW TEST):
* Prepare a stock solution by mixing 1 ml Prostaglandin hormone with 19 ml distilled water.
* Inject the stock solution (1 ml for Cows & 2 ml for Buffaloes) into the ear vein.
* If udder is ready for milk let down within 5 minutes +ve for Pregnancy.
* If there is no milk let down -ve for Pregnancy.
* Care must be taken to remove all the milk from the udder before proceeding for this test.
MPA (MILK PROGESTERONE ASSAY):
* Pregnancy can also be assessed by testing plasma or milk collected from animals after 21-24
days of insemination.
* If the animal is not conceived, the Progesterone hormone will be around 7 mg/ml of milk,
and even lesser in plasma, while the hormone level increases with conception.
PROTEIN BINDING TEST:
* On 24th day of insemination, conception can be known by assessing the progesterone from the
animal’s body hair through protein binding.
PER RECTAL EXAMINATION:
* After 3 months of insemination, the Per rectal examination/palpation of reproductive tract.
