Black Seed Extract: Pest Bioassay Methods

The document outlines a procedure for extracting bioactive compounds from black seed (nigella sativa) using solvents and analyzing them through HPLC and GC-MS. It details the selection and rearing of specific pests under controlled conditions, followed by various bioassays to assess the toxicity and repellency of the black seed extract. Finally, it emphasizes the importance of data analysis, including calculating LC50 and LT50 values and using statistical software for comparison of results.

PEST SELECTION AND REARING

Choose specific pest

Obtain pest from reputable source or establish in house colonies

Mentain pest under controlled conditions ( temp, light, humidity)

BIOASSAY

Contact toxicity bioassay:expose black seed extract to pest using topical application or rumination

Residual toxicity bioassay: evaluate pest mortality after exposure to treated surface I.e leaf disc ,filter
paper

Repellency bioassay: asses pest attraction/repulsion to black treated material

Data analysis

Calculate LC50(lethal conc. 50) and LT50 (lethal time) value

Compare mortality rate between treatment and the control group

Analyse data using statistical software I.e ANOVA

Specific methods

HPLC for phytochemical analysis

GC-MS for volatile compound identification

Bioassay using microplate or petri dish
