History of Microscope
1. Created in 1590
2. Robert Hooke discovered cells using compound microscope
Types of Microscopes
1. Compound Light Microscope*
a. 1st type of microscope
b. Widely used
c. Light passes through 2 lenses
d. Magnification upto 2000x
2. Electron Microscope
a. Used to observe small objects ex. Viruses, dna, parts of cells
b. Uses beams of electrons instead of light
c. More powerful than compound
3. Scanning electron Microscope
a. Can magnify up to 100,000x
4. Transition electron Microscope
a. Can magnify up to 250,000x

Microscope Vocab
*Magnification-increases size of specimen to be viewed under microscope
*Resolution-power to show details clearly
**Both are needed to show clear image
Magnification
3 magnifications: Scanning, low, high
Total magnification= ocular x objective
Magnification Ocular Total Magnification
Lens(eyepiece)
Scanning 4x 10x 40x
Low Power 10x 10x 100x
High Power 40x 10x 400x
Steps to use
Preparation
1. Clear space, backpacks of way
2. Plus microscope into outlet
3. Carry by base and arm with 2 hands
Focusing Specimen
1. Start with scanning objective- Use coarse knob to focus and then use fine knob
2. Once done with scanning, switch to low power- Use coarse knob to refocus, then use fine
knob until clear
3. Finally, switch to high power- Now, only use fine knob to focus
High power(longest objective)
1. Must be focused in low power first
2. Do not use coarse knob
Drawing Specimen
1. Use pencil
2. Include clear and proper labels outside of circle
3. Make large enough to view details
4. Always label specimen name and magnification
5. Specimen should be drawn to scale
Storing
1. Store with scanning objective
2. Wrap cords and cover microscopes
Troubleshooting
Image too dark- Adjust diagram
Spot in viewing field, even when slide is moved- lens is dirty, use lens paper only to clean
Cant see under high power- Go back to lower objectives and adjust
