Practical Title- Isolation of chloroplast

Objectives- To isolate chloroplast and estimate the chlorophyll concentration from


spinach leaves.

Details of First, the animation shows the preparation of a homogenate of plant


tissue, which is obtained by grinding fresh plant material in a buffer

solution. Next, the animation shows the separation of chloroplasts from
other cell components using differential centrifugation. The animation
then demonstrates the washing and purification of the isolated
chloroplasts to remove any contaminants and residual buffer solution.
This is done by resuspending the chloroplast pellet in a fresh buffer
solution and subjecting it to further centrifugation.

The size of the chloroplast in plants varies from 5-10 µm.
