CLONIDINE
Clonidine:Category alpha 2 adrenergic receptor
The treatment of hypertension
Brand name _Catapres
Alpha 2-RECEPTOR
The Apha2 Adrenergic Receptor is an inhibitory
G-protein coupled receptor that binds
norepinephrine and is present in both CNS and
sympathetic arm of the ANS.
Cont's
• with in sympathetic fibers ,alpha2 receptor are
uniquely found on the pre synaptic terminal of
postganglionic.
Mechanism Of Action
• Clonidine treats high blood pressure by
stimulating alpha2 receptor in the brain ,thus
inhibiting the release of norepinephrine ,and
then decreases peripheral vascular resistance,
Cont's
will become vasodilation and the pressure will
decrease.
It has specificity toward presynaptic alpha2
receptor in the vasomotor center in the
brainstem.
Pharmacokinetics
• Bioavailability:immediate release(75-85%)
• Protein bound:20-40%
• Duration:6-10hr
• Metabolism in liver
• Elimination half life:12-16hr
• Excretion in urine
Medical Uses
1. Hypertention
2. Severe dysmenorrhea
3. Vascular migraine headache
4. Treatment of alcohol withdrawal used in
conjunction with benzodiazepines
5. Attention-deficit hyperactivity disorder(ADHD)
6. Restless leg syndrome
7. Tourettes syndrome

Clonidine and Pregnancy
Pregnancy category C
Clonidine may be pass through the barrier of
placenta then enter blood circulation of fetus and
cause decrease blood pressure of the fetus with
decrease heart [Link] can pass into
breast milk and may harm the baby;
contraindicated in pregnant women
Drug-drug Interaction
Asprin
Using asprin
Drug-Food Interactions
Ginseng:may affect the action of the clonidine
and can increase the boold pressure.
