To achieve this unit, the learner must:


1. Perform the different inoculation techniques.
2. Know the use of each manner of inoculation.

MATERIALS USED:
PRE-ANALYTICAL PHASE
1. Inoculating loop/needle must be sterilized over the Bunsen burner flame
1. Prepared Culture Medium
before and after use.
2. Inoculating loops/needle
3. Bunsen burner/ alcohol lamp 2. Sterilization is done by heating the whole length of inoculating loop/needle
up to the distal end of the wire holder but not wire holder till RED HOT while
being held in a vertical position.

3. Allow the heated wire to cool for 5 to 15 seconds before picking up bacterial
colonies. You may allow the heated wire to cool by touching a sterile portion of
the media.

4. After use of any inoculating needle, it should be re-heated (red hot) to


destroy the bacteria adhering to it. If it is covered with viscid material such
as sputum, the inoculating wire should be dried first by holding it near the
side of the flame to avoid spattering and scattering of the living TUBE METHOD
microorganisms.

5. Sterilize all prepared media dispensed into tubes and Petri dishes.

F. POUR PLATE METHOD After the experiment, all the glassware, materials that
have been contact with bacteria like inoculating wire and
including the working station, should be sterilized properly.
The student must prepare 10% bleach or Lysol for
disinfection. Materials like inoculating wire can also be
sterilized by flaming. All disposable materials should be
place properly in correct trash bags.
