Instructions: You can use the “learn” tab to familiarize yourself with the virtual microscope. Click on the “explore” tab to
examine specimens. Your sketches should be drawn as you see them in the field of view. Use the adjustments to get a
clear image.

Sample Slides - Letter E

1. Sketch the view at each magnification.

2. What did you need to do before viewing the slide at 100x?

Switch lens moving through the first three lenses to avoid damaging the [Link]
immersion oil first. Next, clean the lens thoroughly using the correct lens paper.

3. A common mistake for beginners is thinking that the “e” just disappears at the high magnification. How would you
explain to a younger student what happened to the e as you increase the magnification?

The letter "e" did not really disappeared, you just zoomed into microscopic level that you will only
see are its parts:not a whole "e".

Plant Cells

4. Sketch the view at each magnification. You may need to adjust the light.

5. Label Your Drawings: On the 40x view, center the cell, tiny green dots are visible. These are chloroplasts. Label
them on your drawing. You can also see a large, darker circle at this view. That is the nucleus. Label it on your drawing.

Human Blood

6. Sketch the blood cells at each magnification.

7. At 100x, you will see a cell that is larger and has a dark shape in it. This cell doesn’t look like the others because it is a
white blood cell and it functions to protect your body against microbes. Red blood cells do not have a nucleus. White
blood cell count is used as a diagnostic tool to determine if someone is sick. If you were fighting an infection, would you
expect to have more or less WBC’s? Why?

Synthesis and Discussion

8. Compare blood cells to plant cells, include details about color, general shape, and size. Write this as a complete
sentence or paragraph.

Blood cells are generally color red for the red blood cell and white for white-blood cells;on the other hand, plant
cells are green because of the their chlorophyll. Regarding about their shape, blood cells are typically round
shaped and plants cells are typically rectangular or cubic shaped. Lastly, the size of blood cells are smaller
compared to plant cells, but white-blood cell are slightly larger than red blood cell.

Go to the “EXPLORE” tab to view slides. You will need to use the coarse and fine focus and adjust the light to get clear
views of the following. The slide can also be moved within the viewing field.

Sample Slides - Letter E

3. Include a screenshot of the letter “e” at 10x. To take a screenshot, use the print screen on your keyboard or use your
phone to capture the screen. Insert the image in the space below. Your image should clearly show that you viewed the
“e” and focused it.

4. What did you need to do before viewing the slide at 100x?

Switch or change the lens by going through the three lenses before 100x. Wipe the lens using
paper lens and apply immersion oil on the lens.

5. A common mistake for beginners is thinking that the “e” just disappears at the high magnification. How would you
explain to a younger student what happened to the e as you increase the magnification?

It did not really disappear;rather, you just zoomed in it.

Plant Cells

Return to the slide box and choose plant slides → plant cells. Focus with 4x, 10x, and 40x. You may need to adjust the
light and center your slide.

6. Include a screenshot of the plant cells at 40x.

7. Plant cells can be identified by their boxy appearance, where lines represent cell walls. Within the cell are green
circles which are chloroplasts and a dark gray area which represents the nucleus.

At 40x, how many individual cells can be seen in the viewing field? 12

Human Blood

Return to the slide box and choose human → blood Focus with 4x, 10x, and 40x. You may need to adjust the light and
center your slide.

8. Include a screenshot of the blood cells at 40x.

9. Blood cells can be identified by their round appearance. Unlike plant cells, they do not have cell walls.

blood cell count is used as a diagnostic tool to determine if someone is sick. If you were fighting an infection, would you
expect to have more or less WBC’s? Why?

Your WBCs count would tend to go higher when you are sick. Your body must produce or increase more the count of
your WBC to fight off the invader. Note that excessive amount of WBCs is not normal.

Synthesis and Discussion

12. Explore bacteria slides. You don’t need to screenshot them. Compare the bacteria slides to the blood cells and
include details about their color, general shape, and size. Write this as a complete sentence or paragraph.

Bacteria’s shape vary depending if its a rod, sphere, or spiral;while, blood cells are typically
round shape. As for the color and size, bacteria are typically stained purple, most are 0.2 um in
diameter and 2-um length;on the other hand, blood cells are typically round, and are larger than
bacteria.

Common questions

Before viewing at 100x magnification, it is important to use immersion oil between the lens and the slide for clearer imaging. The lens should be cleaned with lens paper, and lenses should be switched methodically without skipping to avoid damage. Proper adjustment of the diaphragm and stage adjustment knobs ensures adequate light and focus .

As magnification increases, finer details of the slide become visible, but the overall view becomes more limited. In the case of the 'letter e', higher magnification doesn't cause it to disappear but instead focuses into such level of detail that only parts of it are visible, not the entire letter .

Kimwipes are not recommended for cleaning microscope lenses as they may cause damage to the delicate surfaces. Instead, lens paper should be used to ensure the lens is maintained without scratches or abrasions .

Under a microscope, red blood cells appear as large round cells usually lacking a nucleus, designed primarily for transporting oxygen. White blood cells are larger with a distinct dark nucleus, and their primary function is to protect the body against infections. Their distinct appearance helps quickly identify them during microscopic examination .

Using lower magnifications before reaching 100x allows the observer to locate and center the specimen more easily while preventing potential damage to the slide or lenses. It provides a broader field of view to identify interesting areas before zooming in for detailed study, ensuring the specimen or microscope isn't damaged during initial focus .

Bacteria can vary in shape, including rods, spheres, and spirals, and are typically stained purple for visibility. They are small, often about 0.2 to 2 micrometers in size. Blood cells, in contrast, are typically round in shape, with red blood cells being larger and generally not requiring staining for identification in microscopy owing to their natural color .

The diaphragm controls the amount of light that passes through the specimen, which is crucial for optimizing visibility and contrast. The stage adjustment knobs move the slide horizontally and vertically, allowing precise positioning of the specimen within the field of view for detailed examination. Both these components are essential for obtaining a clear image .

Immersion oil is used in high-power microscopy, such as at 100x magnification, to increase the resolution of the image by creating a bridge between the lens and the slide. This allows more light to enter the microscope and reduces the refraction of light, resulting in a clearer and more detailed image .

Plant cells are typically rectangular or cubic in shape and green due to the presence of chlorophyll, while blood cells are round in shape. Red blood cells are red in color due to hemoglobin, whereas white blood cells are primarily white. Plant cells are generally larger than blood cells, with white blood cells being slightly larger than red blood cells .

The white blood cell count increases in response to an infection as part of the body's immune response. WBCs are crucial for protecting the body against microbes by multiplying to match the level of threat and attacking the invading pathogens to prevent them from causing harm. This increase serves as a diagnostic tool to indicate the presence of an infection .
