Indirect Methods
• Fonio’s Method
o Materials
▪ Microscopic Slide
▪ Pipette
▪ Diluting Fluid (14% Magnesium Sulfate)
▪ Giemsa or Wright’s Stain
o Procedure
▪ Disinfect the puncture site
▪ After finger puncture, wipe the 1st drop of blood
▪ Place a drop of diluent over the puncture wound
▪ Then press the blood from the puncture site
▪ The ratio of blood to diluting fluid should be 1:3
▪ Make a smear using the mixture
▪ Allow the smears to dry then stain the smear
▪ Count the RBCs and platelets under OIO until 1000 RBCs have been counted
▪ Normal Value = 250,000 – 500,000 per uL
• Dameshek Method
o Materials
▪ Microscopic Slide
▪ Pipette
▪ Pipette Shaker
▪ Rees Ecker Fluid
• Brillant Cresyl Blue (stain) 0.1 gm
• 40% Formalin 0.2 mL
• Sodium Citrate 100 mL
▪ Disinfect the puncture site
▪ After finger puncture, wipe the 1st drop of blood
▪ Place a drop of diluent over the puncture wound
▪ The ratio of blood to diluting fluid should be 1:5
▪ Transfer the mixture into a coverslip and place it on top of a slide
▪ Allow the platelets to settle for 15-45 minutes
▪ Count the RBCs and platelets under OIO until 250 RBCs have been counted
o platelet count/uL = platelet counted x RBC/uL/1000
o Wet mount = count in the center
• Olef’s Method
o Procedure
▪ Disinfect the puncture site
▪ After finger puncture, wipe the 1st drop of blood
▪ Place a drop of diluent over the puncture wound
▪ Then press the blood from the puncture site
▪ The ratio of blood to diluting fluid should be 1:5
▪ Make a smear using the mixture
▪ Allow the smears to dry then stain the smear
▪ Count the RBCs and platelets under OIO until 1000 RBCs have been counted
o Same principle with Fonio’s method
o Confirmation of platelet count can be done on the basis of the occurrence of platelets in
the peripheral smear
o < 1 platelet/OIO field = thrombocytopenia
o 5 – 20 platelets/OIO field = adequate
o > 25 platelet/OIO field = thrombocytosis
• Smear Estimation
o Materials
▪ Microscopic Slide
▪ Microscope
▪ Giemsa or Wright’s Stain
o Procedure
▪ Make a perfect blood smear
▪ Stain with Wright stain
▪ Count platelets in 10 OIO fields
o Platelet estimation = platelet counted x 2000
