Objectives

The literature review threw light upon the present-day condition of the metabolic engineering
tools in the form of Genome scale reconstruction models, metabolic flux analysis and the
more advanced set of tools which integrate the multi-omics data with the GEMs. From the
literature survey the need to address some of the aspects in the modelling techniques have
been listed below in the form of objectives proposed for my project:

1.1. Genome scale reconstruction of Cyanobacteria


Genome scale reconstructed models serve as a very efficient tool in metabolic engineering
and the use of different strains to explore the possibility to create better models is required.
The genome scale reconstruction of such a cyanobacterial species Synechococcus elongatus
PCC 11801 is a proposed goal in the direction to obtain such results.

1.2. High resolution metabolic flux analysis of cyanobacteria


Metabolic flux analysis is the most accurate tool know the flux distribution in the network
because it is based on the experimental data fed to the model. Inferences on the flux
distributions in the above proposed strain of cyanobacteria will help in understanding the
metabolism. This will also pave the way to introduce genetic modifications in the metabolic
network for altering the production of some important metabolite.

1.3. High resolution metabolic flux analysis of engineered strains of


cyanobacteria
The engineered strains of the cyanobacteria will also have its own set of fluxes that may or
may not be as sought with the introduction of the genetic manipulation. The set of flux
distributions so must be estimated and seen to find out the critical points for the irregularities
that will be faced in the mutant organisms.

1.4. Integration of high throughput data in GEM


Incorporation of the multi omics data in the GEM results in better models that have better
predictive performance. The aim will be to obtain a model by integrating the various omics
data with the genome scale model and obtain accurate predictions from the model to more
accurately guide the metabolic engineering process.
