Screening of chitinase producing Bacillus sp.
and antifungal activity against
phytopathogenic fungi
Abstract
Objective: The objective of the present study was aimed to screen of chitinase
producing bacteria and antifungal activity against phytopathogenic fungi.
Methods: Bacteria isolates from marine samples were obtained from the Microbiology
Lab, Science Department, Science and Technology Faculty, Prince of Songkla
University. Bacteria isolates grown by streak plate technique on nutrient agar. All the
bacterial isolates were tested for chitinase activity using colloidal chitin agar media by
spot inoculation method and the potential isolate was used for chitinase production by
submerged fermentation technique using 1% colloidal chitin in the liquid medium. The
efficacy of chitinase activity was assessed by 3,5-dinitrosalicylic acid (DNS) method
along with growth monitoring. The most potential isolate was identified based on
morphological, biochemical, and physiological tests. The most potential Bacillus sp.
B26 isolate was evaluated for its antagonistic activity of crude chitinase against
phytopathogenic fungi namely Aspergillus flavus, Aspergillus niger, Fusarium solani,
and Penicillium chrysogenium used well diffusion method.
Results: Totally, 83 bacteria isolates from marine samples were screened for chitinolytic
activity. Based on the chitinolytic index, 14 isolates showed clear zone hydrolysis on
solid medium. Among the chitinase activity in submerged fermentation, the isolate B26
had prominent chitinase production after 5 days of incubation. The antagonistic activity
of the isolate B26 was effective in restricting the mycelium growth of Fusarium solani
by 44.55 %.
Conclusion: Bacillus sp. B26 has found to possess potent antifungal activity and it can
be used as a promising biocontrol agent against phytopathogenic fungi, thereby
reducing the use of chemical fungicides.
