BLOOD COLLECTION STEPS FOR BACTERIAL CULTURE
1. Under standard precautions
2. Select the site of venipuncture
3. Apply a tourniquet, 3-4 inches above the intended site of venipuncture.
4. Vigorously clean the surface with 70% isopropyl alcohol in a upside down, wait for 30
seconds allow the skin to dry
5. then use 2% chlorhexidine or 10% povidone iodine - beginning in the center and continuing
in an outward direction circularly for an area of 4 to 5 inches in diameter.
6. Allow the povidone iodine to dry ( 1 minute). For chlorhexidine gluconate (2%)-drying period
is 30 seconds. Do not touch the site after cleaning.
7. Alcohol wipe to clean the bottle top
8. Blood is preferably withdrawn from the patient before the giving antibiotics
9. Instruct patient to clench and unclench the fist. Perform phlebotomy using sterile needle
and syringe. Release the tourniquet and withdraw the needle.
10.Blood should be collected in sterile blood culture bottle which contain broth ( Ratio 1: 5 )
11.For Adult -5-10ml. Children - 2-5ml
12.Apply pressure to the site of venipuncture and place a bandage over the puncture site.

Nasopharyngeal swab collection

2. The patient’s head back 75 degrees
3. Gently and slowly insert a swab with a flexible shaft, (plastic) through the nostril parallel to the
palate (not upwards) until resistance is encountered
4. Gently roll the swab
5. Leave the swab in place for several seconds to absorb secretions
6. Slowly remove swab while rotating it
7. Place swab, tip first into the sterile transport tube provided.

URINE COLLECTION STEPS FROM INDWELLING CATHETER

2. the specimen should be collected by clamping the catheter tube for facilitating the
collection of urine above the clamp
3. Then disinfecting the catheter with 70% ethyl alcohol and aspirating the urine using sterile
syringe and needle
4. Transfer the urine in sterile container
5. The urine MUST NOT be collected from the urinary drainage bag

WOUND SWAB COLLECTION STEPS
1. Under standard precautions
2. Cleanse the surrounding skin or mucosal surfaces.
3. For closed wounds disinfect the area with 2% chlorhexidine or 70% alcohol
4. With sterile syringe and needle, pus is aspirated and collected in the sterile container
5. For open wounds, debride the area if needed,
6. Then thoroughly rinse the area with sterile saline prior to collection
7. Then with sterile swab surface swabbing of the infected tissue.
8. Sample collected should have viable infected tissue, rather than superficial debris.
