Microbial Count Protocol for Sugarcane Juice

1. Sample Collection
Collect sample in sterile bottles under aseptic conditions. Store at 4°C and analyze within 1 hour.

2. Total Plate Count (TPC)


Prepare serial dilutions (10^-1 to 10^-4). Take 1 mL sample into sterile petri plates. Add Plate Count
Agar, mix and incubate at 35°C for 48 hours. Count colonies and express as CFU/mL.

3. Total Coliform & Fecal Coliform (MPN)


Inoculate 1 mL sample into lactose broth with Durham tubes. Incubate at 35°C for 24-48 hrs. Gas
formation indicates positive. Confirm using BGB broth (35°C) and EC broth (44°C). Express as
MPN/mL.

4. E. coli Detection
Streak positive EC broth on EMB agar. Incubate at 35°C for 18-24 hrs. Green metallic sheen
colonies indicate E. coli.

5. Yeast and Mold Count


Prepare dilution, take 0.1 mL and spread on PDA. Incubate at 28°C for 48 hrs. Count colonies and
express as CFU/mL.

6. Ice Sample Analysis


Melt ice aseptically and perform same tests: TPC, coliform and E. coli.
