Title: Comparison of microbial growth on different parts of a petri dish

before hand washing and a er washing with water and a er cleaning with
alcohol.

Objec ve:
Compare the different por on of petri dish in different situa ons
1. Without cleaning
2. Washing our hands with soap and water
3. (another person) without cleaning
4. Cleaning with hand sani zer (Like: alcohol)

Materials:
1. Steril petri dish with nutrient agar
2. Marker pen
3. Soap and water
4. Alcohol ( 70 % ethanol )
5. Incubator
6. Gloves

Methods
1. At first, we must divided the petri dish to 5 sec ons by marker
2. Kept the first sec on empty
3. (press your hand slightly in the first sec on without washing )
4. Washing your hand with soap and water, dried, and a finger p was pressed on to the
3rd sec on
5. The next step is handled by second person (finger p was pressed onto 4th part
without cleaning)
6. At the end, Hands were cleaned with alcohol and allowed to dry and pressed onto 5th
sec on
7. The petri dish was covered and put it in incubator at 37C for a 24 h

Observa on and results.


The results show that dirty hands in 2nd and 4th sec ons carry a high number of
microorganisms.
Washing hands with water and soap significantly reduce microbes. But it’s obvious that
some organisms s ll remain.
A er cleaning with alcohol is effec ve but not as much as washing by soap but it kills many
bacteria.

Conclusion: Microbial growth was highest from dirty hands and minimal a er cleaning
with washing with water
And also, it was reduced a er cleaning with alcohol or sani zer.
