Operating Instructions
Recommended Environmental Conditions
In order to extend the machine’s service life, please pay attention to the following factors which may damage
the machine:
• Environmental Conditions;
• Effects of environment, including natural ultraviolet radiation (ie: Direct Sunlight)
• Erosion and abrasion of the protective cover and other safety parts.

• Indoor use;
• Altitude up to 2000m;
• Ambient temperature: +18°C~+30°C;
• Relative humidity: < 80%;
• Operating voltage: 230V, 50/60 Hz;
• Please ensure adequate ventilation in area of use and appropriate sealing measure when performing the
separation of poisonous substances and pathogenic micro-organism cells;
• Ensure there is no vibration and/or a strong draft to influence the performance.
• Ensure there is no dust or flammable and corrosive gas present in the ambient air.
• The setup surface must be firm, level and non-flammable.

Please read this manual carefully when using the instrument for the first time!
The maintenance of the instrument should be carried out by the agents authorized by Biovac.

The following materials should not be used in conjunction with this instrument:

• Flammable and explosive substances;

• Strong chemical materials;
• Toxic or radioactive substances,
• Pathogenic microorganisms

The CO2 Incubator may only be operated by trained, authorized personnel.
Maintenance work on the unit may only be performed by Vacutec Authorised agents.
Tissue, materials or liquids: Which are highly flammable or potentially explosive;

Whose vapours form flammable or explosive mixtures with air;
Which release toxins, May not be used!
The pressure of the CO2 Supply can be adjusted to a range of 0,8-1 bar.
CO2 is a gas which may pose as a potential heath hazard, therefore the unit should be serviced at regular
intervals.

Only qualified personnel with specialised equipment may work on supply lines and compressed gas
container, bottles or collective systems in which CO2 is stored for use with the incubator.

3. Application
The Gassed Incubator can simulate the natural physiological parameters of cell and tissue cultures.

The unit must not be used for treatment of flammable materials that may explode.

4. Abbreviated operating instructions


Factory Settings: Temp. ––– 37.0ºC, %CO2 ––– 0%
Each time at Startup, the following instructions Must be strictly complied to:

a. Open the glass door, fill the reservoir with distilled water, close the door.
b. Turn the “Over-Temp” control knob to 39ºC
c. Connect the gas interface of the unit to the CO2 cylinder.
d. Ensure the power switch at the rear of the unit is “Off ”.
e. Connect the unit to the power supply and turn on the power.
f. The unit will automatically conduct a routine self-test while displaying [%CO2] and [ºC] as [888].
g. Self-test lasts roughly 40 seconds, after which the [%CO2] and [ºC] will display the actual values measured
inside the chamber.
! h. Press [ ] and [ ] together for about 10 seconds, until [%CO2] shows as [0.0] and the “Auto-start” light
comes on, then release both buttons.
i. The unit will begin the “Auto-start” function.
j. Allow the Incubator to stand for 16-24 hours. !

k. Once the Temp. of the unit displays 37.0 ± 0.1ºC, and CO2 displays 0 ± 0.1% CO2, Press !and hold [ ] key

5. Description of the Unit


5.1 Structure WARNING:

5.1.3 The inner casing is made of stainless steel.

5.2 Internal Fittings:


The pullout shelves are tilt-proof and adjustable over a height of 50mm. Removal of shelves and carrier
requires no tools and can be removed simultaneously.

5.3 Heating System:


5.3.1 Large-surface heating cables are arranged on the outside of the inner casing, to heat the interior
space. The heating cables are placed beneath the water reservoir, on the ceiling , on the upper part of the
rear wall and in front of the outer door.
5.3.2 Due to the outer door being heated, condensation will not precipitate on the glass door. Therefore
the interior remains clearly visible despite the high humidity. Condensation may form if the outer door is
left open for prolonged periods.

5.3.3 The new normal / slow-heating mode enables the incubator to work properly in different Temp.
conditions. Normal heating mode is recommended.

The special heating system can efficiently operate on 8ºC above ambient Temp.
If the unit is to be operated at lower Temp., you can turn off door heating system and the unit can
operate at 5ºC above ambient Temp. (refer to section 9.7 Special Function 5: Door Heating system).
In this instance, you may experience a little condensation on the corner of the glass door. During
normal operation, the door heating system is “ON” and the auxiliary heating is operating.

5.4 Humidity
5.4.1 The distilled water kept in the reservoir of the unit, evaporates and humidifies the working chamber.
Under normal operating conditions the relative humidity inside the chamber will be ≥ 95%

5.4.2 Because of teh unique heating system, condensation will not precipitate on the chamber ceiling and
glass door. The sidewalls and the rear wall of the chamber serve as appointed condensation surfaces.

5.4.3 Water can be removed from the basin using a pump
a. Prepare teh pipette for removing the water.
b. Remove the shelf inside the incubator.

c. Connect one side of the pipette with the output of the electric siphon pump.
d. Press the pump with four suction cups onto the recess at the rear, inside the incubator with the power
cable extending from the top of the pump and plugged into the required power supply.
e. Drain the water until all is removed.
f. Turn off the pump and remove it from the incubator
g. Wipe down the pump and reservoir.

5.5 Gas Supply:


A suitable connector for the CO2 supply is
located on the rear wall of the unit (figure 3)

Please observe the markings of the CO2 connector!

Before entering the chamber, the gas passes a sterile filter where particles larger than 0.3um are retained.
Filter efficiency is 99.998%. A small fan ensures that the gas and in incubator atmosphere are thoroughly
mixed.

5.6 Door Switch:
There is a door switch located behind the glass door. When the glass door is opened, the switch cuts off
the gas supply and the heating system in order to avoid uncontrolled admission of gas or heating of the
chamber. With the glass door open, all displays will flash until the door is shut again. An alarm will be
activated if the door remains open for more than 5 minutes. The outer door can only be closed once the
glass door is locked and air proofed.

5.7 Pressure Compensation:


A pressure compensation vent in the lower section of the chamber rear wall avoids undesirable pressure
build-up during admission of gas. This vent is connected with an overflow (Unique direction) valve on
the rear wall of the unit. The valve impedes unwanted flow of ambient air when the glass door is open.
(figure 4)

6: Microprocessor Controlled System

6.1 Physical parameter Control:


6.1.1 Temp:
The chamber Temp. is precisely regulated by a microprocessor-based control circuit. Platinum Pt 1000
resistance serve as Temp. sensor. An over-Temp. sensor will automatically cut off the heating system and
trigger an alarm.

6.1.2 CO2 Control: N


a. In order to keep the PH value of the culture media constant at a set value, you need to control the CO2
concentration of the chamber atmosphere. The relevant CO2 concentration depends on the desired PH
value and the content of sodium bicarbonate buffer in the medium.
b. CO2 measurement is based on continuous measurement ofWARNING:the thermal conductivity level of

6.2 Automatic Calibration of the Measuring System:


AUTO-START Routine
6.2.1 Start-up the unit or changing the Temp. set point requires calibration of the various measuring system.
Your incubator performs this task automatically during the AUTO-START routine.

6.2.2 When both temp. and humidityhave reached teh steady state, the microprocessor recognizes and
performs a fully automatic calibration of the measuring system.

6.2.3 When the Temp. is equals to the ambient, this process requires approx. 15 hours. Prior to AUTO-
START, make sure that the chamber is void of all gases, except air.

ADVICE: You should perform an AUTO-START once every 6 weeks.

6.3 Monitoring Functions:


6.3.1 Door Control:
All actual value displays will flash as soon as the outer door is opened and will continue to do so until
the door is properly closed again. Should the door remain open for more than 5 minutes, the unit will
generate an error message and relative error code can be read.

6.3.2 Error Recognition:
A microprocessor reliably monitors all chamber atmosphere parameters and detects any malfunctions
that might occur. All set values have definite tolerance ranges and an error message will be triggered in
case the actual value is outside the tolerance range.
6.3.3 Tolerance Range
- Temp.: ± 0.5ºC
- CO2 content: ± 1.0% CO2

6.3.4 Error Messages:


a. Errors detected by the microprocessor are assigned to special code numbers. These code numbers will
assist you in identifying the malfunction.
b. When control circuit or displays are affected (e.g. door is open), the LED will come on to remind you
of the malfuntion in the unit. You will hear an alarm sound if the buzzer is connected. Press [i] key to
view the error message. The displays will show [ ––– ] if there are no errors.

6.3.5 Alarm:

If a malfunction occures, you should first press the [i] key to view the error message.
The error message will be deleted if:
• The Set point changes
• You turn the unit off / on.

6.3.6 Error Code Table:

6.4 Special Functions:


For adjustment, refer to section 9.7
Use the Function Key, identified by a [ i ] symbol, to select the special functions below.

6.4.1 Buzzer On/Off:


• Function 1:
If the buzzer is turned on you will get an accoustic alarm when errors occur.
Buzzer Off: “Silent” alarm.
Factory Setting: Buzzer ON.

6.4.2 Gas On/Off:


• Function 2:
With the selection of “Gass Off ”, the gas supply is cut off permanently, the green “Gas” LED is illiminated.
Factory Setting: Gas ON, the green “Gas” LED will come on.

6.4.3 CO2 zero-calibration:
• Function 3:
In case the CO2 deviation is too large, this function will assist you in calibrating the CO2 zero point.

6.4.4 Set point Lock-in:


• Function 4:
This function permits you to lock the set points you have entered for Temp. and CO2 so that these values
cannot be changed inadvertently.
Factory Setting: Set point unlocked.

6.4.5 Door heating system on/off


• Function 5:
Normally, the working Temp. is 8ºC higher than the ambient Temp. If the unit is to be operated at lower
Temp., it can be used at only 5ºC above ambient by turning off the “soft” switch in function 5. In this
instance, you might experience a little condensation on the glass door.

6.4.6 Heating mode Normal / Slow


• Function 6:
It provides tow heating modes: normal and slow. This enables the indicator to work well under different
Temp. conditions. Slow mode is usually used under high ambient Temp. while Normal heating mode is
recommended.

6.4.7 Operating Modes: With / Without Water


• Function 7:
If you want to run your unit without water in the reservoir, you need to select this mode prior the start-up
Factory Setting: To be run with water in the reservoir.

7. Over-Temp Protection

7.1 Temp. Limit Controller - (Responds to Temp.)

7.1.1 The unit features an independent electronic Temp. limit controller to protect the incubator, it prevents
the working chamber from over heating. (refer to figure 1 - In compliance with the Thermal Safety Class
3.1 in accordance with DIN 12 880).

7.1.2 In case of a Temp control circuit malfunction where the set Temp is exceeded, the Temp. limit
controller will take over the control function.

7.1.3 Setting it requires a tool (a coin or screwdriver, for example); refer to Section 9.5

7.2 Protecting the unit:


If you set the control knob to 55ºC (limit stop) the Temp. limit controller will protect the incubator.

7.3 Protecting Culture:


7.3.1 The Temp. limit controller will protect teh culture if you set it approx 1-2ºC higher than the Temp.

7.3.2 The red indicator will illuminate as soon as the Temp. limit controller responds.

7.3.3 In the case where the Temp. limit controller has taken over the control function, refer to 10.1 Error
Code Table to investigate and determine the cause of the malfunction.

7.4 Operational Test:

At resonable intervals your Temp. limit controller should be tested for proper operation. To run this test, turn
the knob to where is shows approx. 2ºC less than the Temp. indicated on the display. The red indicator
light should come on. We recommend that your authorised operators run this test on a routine basis, e.g.
Prior to a lengthy working process.

8: Location and Installation

8.1 Location
a. Unpack your unit and remove all accessories from the interior.
b It must be located in a dry area where the ambient temperature does not exceed 30ºC
c. Place the unit on a level, non-combustible surface that does not vibrate.
d. The gassed incubator will run with CO2, which is not harmful to the operator when the room is
adequately ventilated.. The gases escape from the back wall of the unit, therefore adequate space must be
allowed around the unit to ensure the gas emmissions are not restricted.
e. The unit should not be placed and operated in areas without adequate ventilation
f. If several units are placed in the same room or the unit is installed under a laboratory bench, additional
ventilation equipment must be installed.
g. For information on gases released during operation, consult the Appendix.

8.2 Postioning Distances:


To ensure adequate ventilation, the required space surrounding the unit should be no less than 50mm left,
right and top, while no less than 120mm between the rear of the unit and any other object or wall.

DO NOT block the overflow valve on the rear wall of the unit. Do not connect a hose to the valve outlet as
this could cause condensation to occur.

8.3 Stacking:
Refer to section 13.1

8.4 Installation, Connection to Gas Supply:


a. The appropriate interface is placed on teh back of the unit. A relative hose is provided with the unit. The
gas required for operation of the unit is to be connected via a pressure reduction valve, set a 1 bar. The
pressure reduction valve will only be supplied on customer’s request.
b. For technical safety reasons, the 1 bar setting must not be changed.

8.5 CO2:
Connect the gas cylinder, with the pressure reduction valve, to the unit.

8.6 Reminder:

The gases must be at least 99.5% pure.


Do not use a CO2 cylinder with a cart.

8.7 Connection to Main Power Supply:
a. Prior to connecting the unit to mains, ensure that the power ratings match those stated on the nameplate.
b. Your gased incubator is equipped with a properly grounded power cable.
c. A 6.3 delay-action type fuse is required for mains fusing.

9. Start-Up
The ambient Temp. of the location must be at least 8ºC lower than the Temp. setting. Keep the outer door
and the glass door open.

9.6 AUTO-START
9.6.1 ATTENTION! the chamber must be void of any gases except air.

11.4 Maintenance
It is recommended to run an AUTO-START at least once every 2 months to re-calibrate the gas analyzer.

We only guarantee safety of the unit when maintenance, repairing or


modifications are conducted by personnel authorized by Vacutec.
Check your unit at regular intervals to ensure proper operation.
The unit should be inspected anually for safety, leak proof and a service.

11.5 UV light
In operation, fungi may develope in the incubator. To protect the unit from contamination and keep
samples pure in the chamber, a UV light can be applied to disinfect the inner chamber.

11.5.1 Location of UV Light


UV light is located on the upper part of the rear wall of the inner case. It is connected to the
microprocessor controller through two special, air tight, water proof, Silastic lamp sockets. The protection

switch (UV light switch) is positioned on the back wall of the incubator and is marked as (UV). Please
keep the switch in off position to avoid accidental triggering of UV light which may destroy samples.
After long periods of use, the lamp may turn black at each end, burning out. In this case, the UV bulb will
need replacing.

DO NOT CHANGE THE UV BULB YOURSELF, this procedure may only be performed by an authorised
Vacutec Service Engineer.

11.5.2 How to use the UV light


If you want to turn on the UV light, two switches need to be activated. One switch is a software switch,
Function 222, the other is a hardware switch found on the rear wall of the incubator.
Steps:
1. At start up, the incubator will perform a routine self-test for roughly 40 seconds. [888] displayed on the
panel will then reflect an actual value.

2. Before starting the UV light disinfecting function, ensure that there are no samples in the incubator!

13 Appendix

13.1 Stacking of Gassed Incubators:

If several units are placed in the same room or if a unit is installed underneath a laboratory bench,
additional ventilation equipment must be provided.

a. One gassed incubator can be placed on top of another with the provided stacking feet.
b. One set includes four stacking feet onto which the upper unit is fixed
c. The 4 stacking feet should be screwed into the holes on the upper part of the unit with the enclosed
self-tapping screws.

13.2 Gas Volume for Normal Operation and Malfunction


Explanation of terms used:
1. Required amount of gas: The amount of gas needed in order to reach the max adjustable set point.
2. Gas consumption: The volume of gas constantly injected to maintain the max adjustable set point.
3. Malfunction: When over pressurisation occurs due the gas exceeding the maximum set point, an
audible alarm will sound accompanied by a visual alarm.
4. Gas flow: With the valve fully open, the volume of gas flowing through the capillaries per hour.
